API Examples
Atacama Labs Oy has developed a number of formulations, both for in-house programmes and for external clients. In the programmes, Atacama Labs Oy has granulated various excipients and combinations of APIs in order to:
In the internal trials, Atacama Labs Oy has processed various pharmaceutical substances with different characteristics. Below, see details of Paracetamol, Ibuprofen, Metformin and Avicel trials. The other examples include substances like Sumatriptan, Ketoprofen, Fluoxetine, Acebutolol, L-arginine, and Nicorandil.
Granules with promising flow properties and compressibility have been produced in each case. Take a quick tour through these examples, you may have seen some of these problems now overcome earlier in your career. Looking back at these successful examples we continue our efforts in the future with our development program including representative examples for known development difficulties.
With PDG Technology™, paracetamol can be granulated on its own (100%) or with various excipients. Atacama Labs has achieved acceptable hardness and disintegration properties. With PDG Technology™, the result of the granulation was a well flowing granulate and the tablets with high drug-loading (90.9%) that are strong enough (104N), disintegration time less than 15 minutes.

With PDG Technology™, Metformin is very easy to granulate. The Metformin granules show excellent flow properties, are dust free and porous. Click here to download a Metformin granulation and tabletting report.
The evident result was that the PDG granules have excellent flowability while the loss in compactibility is acceptable.

Paracetamol
Paracetamol/acetaminophen is known to be a difficult material to dry granulate. The majority, if not all, commercial products are produced using the wet granulation processes. With PDG Technology™, paracetamol can be granulated on its own (100%) or with various excipients. Atacama Labs has achieved acceptable hardness and disintegration properties. Ibuprofen
Ibuprofen is a difficult-to-process material due to its stickiness and low melting point. With PDG Technology™, the result of the granulation was a well flowing granulate and the tablets with high drug-loading (90.9%) that are strong enough (104N), disintegration time less than 15 minutes.

Metformin hydrochloride
Metformin hydrochloride is a biguanide oral hypoglycaemic agent. The raw material is known to be relatively difficult to dry granulate due to the fact that it is hygroscopic, cohesive / “sticky”, it has stability problems, and it is not inherently compressible. With PDG Technology™, Metformin is very easy to granulate. The Metformin granules show excellent flow properties, are dust free and porous. Click here to download a Metformin granulation and tabletting report.
Avicel
Roller compacted Avicel (PH105) was compared to PDG granulated Avicel (PH105). The roller compacted material was processed with 5 and 11 KN compaction forces (in order to achieve flowability) while the PDG granulate was processed with only 1KN compaction force. The evident result was that the PDG granules have excellent flowability while the loss in compactibility is acceptable.
